Recently, the Kunming Institute of Zoology, Chinese Academy of Sciences ion channel drug research and development center and the Shanghai Long March Hospital, for the first time carried out in line with international standards of borneol clinical research, explained the traditional Chinese medicine borneol external analgesic mechanism, to participate in the total number of clinical trials 122 people.
Borneol is a common Chinese herbal medicine, Chinese medicine clinical application more than a thousand years. Natural borneol is an almost pure organic compound obtained from the Lauraceae plant. Its chemical composition is (+) - Borneol, which belongs to the small molecule terpenes. Borneol "taste hard, cool", it is an important effect is "swelling and pain", can be used for "sore swelling and pain" "limb pain" and so on. However, the clinical efficacy of topical borneol analgesia has been lacking in human trials that meet international standards (randomized, placebo-controlled, and double-blind), and the scientific mechanisms of analgesia have not been elucidated. For a long time, the US Food and Drug Administration has set the borneol as a food additive and is not listed as a drug.
The researchers conducted randomized, randomized, placebo-controlled, double-blind clinical trials to demonstrate that topical borneol had a good analgesic effect on postoperative patient wounds. The TRPM8 ion channel in peripheral neurons was the most sensitive target for borneol in mammals, so far, by screening the different molecular targets on the pain signaling pathway. By further establishing the mouse pain model, knockout mouse TRPM8 gene and pharmacological blocking test, proved that the analgesic effect of external borneol is mainly mediated by TRPM8 channel. Borneol activates TRPM8 peripheral neurons, which in turn activates presynaptic glutamate receptors in the dorsal horn of the spinal cord, leading to inhibition of pain nerve signaling. For the first time, this study provides clinical data of topical icole analgesia in line with international standards, and elucidates the mechanism of cell molecular mechanism, which is of great significance to the modernization and development of traditional Chinese medicine borneol.
